The alleged incident, which took place in Albuquerque, New Mexico, happened as the young girl was travelling with her parents along the Interstate 40.
Police said that a car drove up alongside the family's vehicle before someone from inside it began
firing bullets.
Both parents were unharmed, however the four-year-old girl was shot in the head.
The suspect then fled the scene.
The girl - who has not been named - was rushed to hospital following the tragic incident, where she later died.
Police are now urging any witnesses to come forward after having received conflicting information about the suspect's vehicle. Albuquerque Police Department Chief Gorden Eden said: "We are in desperate need of help to find this suspect.
"It's a terrible, tragic loss, and it shouldn't have ever happened.
"This is one of those crimes which is unexplainable. There is no way to explain your way out of this."
